The Phase III results of the Lp(a) HORIZON trial for pelacarsen have been announced. The trial has failed to reach its primary endpoint: although Lp(a) levels were lowered, this did not result in a statistically significant reduction in cardiovascular events. Similar results were observed in the Phase III ZEUS trial of ziltivekimab, which approached residual CV risk from a different direction, Although ziltivekimab successfully inhibited the IL-6 pathway, it did not reduce CV risk. These trial results further highlight the fact that the relationship between biomarkers with the increased risk of disease is complex. The quest for molecules that represent therapeutically modifiable cause of disease continues.
